Story Line:  John Strangways, the MI6 Station Chief in Jamaica, and his secretary Mary, meet a grim fate at the hands of a trio of assassins who also ransack Strangways' residence. Learning of Strangways' demise, M, the head of MI6, calls upon intelligence officer James Bond to investigate the incident, especially to determine if it relates to Strangways' collaboration with the CIA on a case linked to radio jamming disrupting rocket launches at Cape Canaveral. Upon Bond's arrival in Jamaica, an apparent chauffeur, who is an enemy agent with deadly intentions, attempts to eliminate him but falls prey to Bond's resourcefulness. Unfortunately, the agent takes his own life with a cyanide-laced cigarette before Bond can interrogate him.


At government headquarters, Bond questions Pleydell Smith about Dent's involvement. Subsequently, at Strangways' residence, Bond confronts a local boatman named Quarrel, who has CIA ties and introduces Bond to their agent Felix Leiter, also investigating Strangways' death.


Bond discovers from Felix that the CIA traced the radio jamming signal to Jamaica, and Strangways was helping pinpoint its source. Quarrel reveals they collected mineral samples from the forbidden island of Crab Key before Strangways' demise. Upon finding a receipt from a local geologist, Professor R. J. Dent, Bond's suspicions grow as Dent's explanations seem dubious. Dent visits Crab Key's reclusive owner as instructed, hoping to have Bond killed by a tarantula, but Bond prevails and sets a trap. He turns the tables on the assassins known as the "three blind mice" and has Miss Taro, Pleydell Smith's unscrupulous secretary, arrested.


Bond, suspecting that Dent was instructed to test Strangways' samples for radioactivity, confronts Dent, who doesn't survive the encounter. After checking Quarrel's boat with a Geiger counter, Bond deduces that Strangways suspected Crab Key was the source of the radio jamming and persuades Quarrel to take him there. Upon their arrival, Bond encounters Honey Ryder, a shell diver, and escapes armed guards with her and Quarrel into the swamp. They are captured and taken to a hidden base where they are decontaminated due to radiation in the swamp and later rendered unconscious with drugged coffee.


Upon waking, they meet the base's owner, Dr. Julius No, a Chinese-German criminal scientist with metal hands due to radiation exposure. Bond learns that No was once part of a Chinese crime organization and now works for SPECTRE, planning to use the radio jamming to disrupt the Project Mercury space launch at Cape Canaveral as a display of SPECTRE's might. Bond refuses to join SPECTRE, leading to Ryder's separation and Bond's imprisonment. Bond escapes his cell through an air vent, infiltrates the base's control center, and sabotages the nuclear pool reactor powering the radio beam, preventing the launch disruption. Dr. No tries to stop him but meets a gruesome end in the reactor pool.


As the base evacuates, Bond liberates Ryder, and they escape the island by boat just before the base's destruction. Felix finds them at sea, and a Royal Navy ship tows them to safety. Bond releases the tow rope to embrace Ryder as she passionately kisses him.
